    睡眠を司るホルモンであるメラトニンは脳(松果体)で合成・分泌されるホルモンで、自律神経(交感神経)の活動により分泌調節される。臨床的には、皮膚をさする刺激が入眠を促し、睡眠の質を高めることが報告されているが、皮膚への刺激がメラトニン分泌に影響を及ぼすことを実証した報告は見当たらない。そこで本研究では、皮膚への刺激が松果体からのメラトニン分泌に及ぼす影響とその神経性機序を明らかにすることを目的とする。 今年度は、マイクロダイアリシス法によりラットの松果体の潅流液を回収し、液中に含まれるメラトニン量をELISA法により定量化する方法を確立した。 麻酔下の雄性ラットの頭蓋骨の一部を除去して上矢状静脈を切除し、松果体を視認しながら透析膜のついたマイクロダイアリシスプローブを刺入した。灌流液を毎分1μLの速度で流し、氷上に置いたサンプルカップ中に灌流液を回収した。 その結果、回収した灌流液中に含まれるメラトニン濃度は数10 pg/mL程度であること、β受容体刺激薬を全身投与することにより灌流液中のメラトニン濃度が著しく増加することが分かった。 さらに、灌流液を20分毎に繰り返し採取し灌流液中のメラトニン濃度の経時変化を検討した。その結果、灌流液中のメラトニン濃度は安定しており、サンプル間の違いは約10%程度であることが分かった。

    Musculoskeletal disorders such as neck-shoulder stiffness and low back pain are one of chronic symptoms that many people suffer from. It has been reported that patients with chronic pain may also have cardiovascular dysfunctions such as orthostatic hypotension. The present study aimed to elucidate plastic change of cardiovascular response to skeletal muscle pain and its neural mechanisms, particularly focusing on inflammation which is related to pain chronicity. Experiments were performed on anesthetized rats. To induce inflammation in skeletal muscles, carrageenan solution was intramuscularly injected a day before experiment. Pressure stimulation applied to rat's calf altered heart rate and blood pressure. When pressure stimulation was applied to an inflamed calf, increases in heart rate and blood pressure were exaggerated. Additionally, the contribution of cardiac sympathetic efferent nerves to such cardiovascular responses was suggested.

    Mechanical pressure stimulation applied to the calf alters heart rate in anesthetized animals. The aim of the present study was to establish an assessment method for muscle pain in anesthetized animals by using cardiac function changes in response to muscle stimulation. Heart rate, arterial pressure and cardiac sympathetic nerve activity were recorded during experiments. Heart rate changes (increase and decrease) in response to pressure stimulation of the calf muscles were almost abolished by blocking cardiac sympathetic nerve activity. Additionally, experiments that cardiac sympathetic nerve activity were recorded revealed that nerve activity changes occurred in parallel with heart rate changes. In the calf inflamed by inflammatory substance administration, pressure stimulation-induced increases in cardiac sympathetic nerve activity and heart rate exaggerated.

    The present study examined the effect of gentle cutaneous stimulation (touch) and its mechanism using heart rate responses to noxious heat stimulation as an indicator in anesthetized rats. We found that touch using a soft elastomer brush inhibited heat-induced heart rate responses. Additionally, the inhibitory effect of touch on heart rate responses was abolished by blocking spinal μ-opioid receptors.
